FUNCTION: Catalyzes the decarboxylative condensation of pimeloyl-CoA and L-alanine to produce 8-amino-7-oxononanoate (AON), coenzyme A, and carbon dioxide (By similarity).
CATALYTIC ACTIVITY: 6-carboxyhexanoyl-CoA + L-alanine = 8-amino-7-oxononanoate + CoA + CO(2).
COFACTOR: Pyridoxal phosphate (By similarity).
PATHWAY: Cofactor biosynthesis; biotin biosynthesis; 8-amino-7-oxononanoate from pimeloyl-CoA: step 1/1.
SUBUNIT: Homodimer (By similarity).
SIMILARITY: Belongs to the class-II pyridoxal-phosphate-dependent aminotransferase family. BioF subfamily.
GO:0008710; Molecular function: 8-amino-7-oxononanoate synthase activity.
GO:0030170; Molecular function: pyridoxal phosphate binding.
GO:0009102; Biological process: biotin biosynthetic process.

Biof (8-amino-7-oxononanoate synthase)(TIGR00858) and kbl (2-amino-3-ketobutyrate coenzyme A ligase)(TIGR01822) catalyze very close reactions, and alignment of the amino acid sequences of both enzymes reveals a very close homology, which does not allow to separate them. The only way to create a specific family to the bioF enzymes is to limit the rule to the Proteobacteria. A few Firmicutes (LYSSH and some close orthologs), are annotated as a 8-amino-7-oxononanoate synthase (bioF) due to the characterisation but are not members of the family. The rest of proteins which cannot be differentiates, is annotated as a putative 8-amino-7-oxononanoate synthase ligase in DE line and are not members of the family. Proteins with both activites (TIGR00858/TIGR01825) are annotated as putative 8-amino-7-oxononanoate synthase/2-amino-3-ketobutyrate coenzyme A ligase in DE lines and BIKB in ID lines. |
