case <OC:Mammalia>
Function Component of the transcription regulatory histone acetylation (HAT) complex SAGA, a multiprotein complex that activates transcription by remodeling chromatin and mediating histone acetylation and deubiquitination. Within the SAGA complex, participates in a subcomplex that specifically deubiquitinates both histones H2A and H2B. The SAGA complex is recruited to specific gene promoters by activators such as @gn(MYC), where it is required for transcription. Required for nuclear receptor-mediated transactivation. Within the complex, it is required to recruit USP22 and ENY2 into the SAGA complex. Regulates H2B monoubiquitination (H2Bub1) levels. Affects subcellular distribution of ENY2, USP22 and ATXN7L3B.
Subunit Component of some SAGA transcription coactivator-HAT complexes, at least composed of ATXN7, ATXN7L3, ENY2, GCN5L2, SUPT3H, TAF10, TRRAP and USP22. Within the SAGA complex, ENY2, ATXN7, ATXN7L3, and USP22 form an additional subcomplex of SAGA called the DUB module (deubiquitination module). Interacts directly with ENY2 and USP22.
Domain The C-terminal SGF11-type zinc-finger domain together with the C-terminal catalytic domain of @gn(USP22) forms the 'catalytic lobe' of the SAGA deubiquitination module.
else case <OC:Drosophilidae>
Function Component of the transcription regulatory histone acetylation (HAT) complex SAGA, a multiprotein complex that activates transcription by remodeling chromatin and mediating histone acetylation and deubiquitination. Within the SAGA complex, participates in a subcomplex that specifically deubiquitinates histone H2B. The SAGA complex is recruited to specific gene promoters by activators, where it is required for transcription. Required for nuclear receptor-mediated transactivation. Binds independently on SAGA to promoters in an RNA-dependent manner. Binds to mRNA and is essential for total mRNA export from the nucleus. Required to counteract heterochromatin silencing. Controls the development of neuronal connectivity in visual system by being required for accurate axon targeting in the optic lobe. Required for expression of ecdysone-induced genes such as br/broad.
Subunit Component of some SAGA transcription coactivator-HAT complexes, at least composed of Ada2b, not/nonstop, Pcaf/Gcn5, Sgf11 and Spt3. Within the SAGA complex, Sgf11, e(y)2, and not/nonstop form an additional subcomplex of SAGA called the DUB module (deubiquitination module). Interacts directly with not/nonstop. Interacts with the AMEX complex component xmas-2. Interacts with Cbp80; important for promoter recruitment of Sgf11 that is not associated with the DUB module.
Domain The C-terminal SGF11-type zinc-finger domain together with the C-terminal catalytic domain of not/nonstop forms the 'catalytic lobe' of the SAGA deubiquitination module.
else case <OC:Fungi>
Function Functions as component of the transcription regulatory histone acetylation (HAT) complex SAGA. At the promoters, SAGA is required for recruitment of the basal transcription machinery. It influences RNA polymerase II transcriptional activity through different activities such as TBP interaction and promoter selectivity, interaction with transcription activators, and chromatin modification through histone acetylation and deubiquitination. SAGA acetylates nucleosomal histone H3 to some extent (to form H3K9ac, H3K14ac, H3K18ac and H3K23ac). SAGA interacts with DNA via upstream activating sequences (UASs). Involved in transcriptional regulation of a subset of SAGA-regulated genes. Within the SAGA complex, participates in a subcomplex, that specifically deubiquitinates histones H2B.
Subunit Component of the 1.8 MDa SAGA transcription coactivator-HAT complex. SAGA is built of 5 distinct domains with specialized functions. Within the SAGA complex, @gn(SUS1), @gn(SGF11), @gn(SGF73) and @gn(UBP8) form an additional subcomplex of SAGA called the DUB module (deubiquitination module). Interacts directly with @gn(SGF73), @gn(SUS1) and @gn(UBP8).
Domain The C-terminal SGF11-type zinc-finger domain together with the C-terminal catalytic domain of @gn(UBP8) forms the 'catalytic lobe' of the SAGA deubiquitination module.
